Beyond electron transfer, NAD+ serves as a substrate for enzymes that cleave the molecule and transfer ADP-ribose units. Sirtuins, poly(ADP-ribose) polymerases, and CD38 consume NAD+ in regulatory reactions. These activities link NAD+ availability to DNA repair, chromatin modification, calcium signaling, and metabolic stress responses. Because consumption can exceed biosynthesis under some conditions, cellular NAD+ levels are dynamic rather than fixed. Enzyme affinity and local synthesis also influence how much NAD+ is available for signaling.
NAD+ is a dinucleotide composed of adenine, ribose, and nicotinamide moieties linked by phosphate groups. Its oxidized form carries a positive charge on the nicotinamide ring, which enables reversible hydride transfer. The molecule functions as a coenzyme in oxidoreductase reactions rather than as a dietary vitamin in its intact form. Cells maintain separate pools in cytoplasm, mitochondria, and nucleus. This compartmentalization allows distinct redox environments while preserving a shared chemical identity.
Beyond redox chemistry, NAD+ serves as a substrate for enzymes that cleave the molecule and transfer its ADP-ribose moiety or remove acetyl groups. Sirtuins consume NAD+ during deacetylation, poly(ADP-ribose) polymerases use it in DNA damage responses, and CD38 enzymes hydrolyze it to signaling metabolites. These consumption pathways mean that NAD+ availability can influence gene regulation, DNA repair, and calcium signaling. Cellular NAD+ concentrations decline in some tissues with age in animal models, but whether this decline is a cause or consequence of aging in humans remains an active open question.
Nicotinamide adenine dinucleotide, commonly abbreviated NAD+, is a dinucleotide coenzyme built from an adenine nucleotide and a nicotinamide nucleotide joined by a pyrophosphate linkage. Its oxidized form carries a positive charge on the nicotinamide ring, while the reduced form, NADH, carries a hydride equivalent. The molecule participates in hundreds of oxidoreductase reactions, where it accepts or donates electrons and protons. Because it can cycle between oxidized and reduced states without net consumption, NAD+ functions as a reusable electron carrier rather than a fuel molecule.
In glycolysis, the tricarboxylic acid cycle, and fatty acid oxidation, NAD+ is reduced to NADH at specific dehydrogenase steps. NADH then delivers electrons to the mitochondrial electron transport chain, mainly at complex I, supporting oxidative phosphorylation and ATP production. The balance between NAD+ and NADH, often expressed as a ratio, influences metabolic flux and redox homeostasis in different cellular compartments. Cytosolic and mitochondrial pools are connected but not identical, and their ratios can differ substantially because of compartment-specific enzymes and transport systems.

NAD+ also serves as a substrate for enzymes that cleave it, including sirtuins, PARPs, and CD38. These enzymes consume NAD+ and release nicotinamide and ADP-ribose or related products. The dual roles as redox cofactor and signaling substrate connect NAD+ to DNA repair, circadian regulation, and calcium signaling. Cellular NAD+ concentrations vary by tissue, time of day, and stress exposure. How these consumption pathways interact with redox balance remains an active area of research.
NAD+ is a dinucleotide composed of two nucleotides joined by a pyrophosphate linkage. One nucleotide contains adenine, and the other contains nicotinamide. The oxidized form carries a positive charge on the nicotinamide ring and is abbreviated NAD+. It functions as a cofactor in hydride-transfer reactions, accepting electrons in catabolic pathways. In cells, it interconverts with reduced NADH, forming a redox couple central to energy metabolism. The molecule is water-soluble and does not cross cell membranes freely without specific transport or precursor pathways.
The nicotinamide ring undergoes reversible reduction at the para position, converting NAD+ to NADH. This reaction transfers a hydride equivalent, not a free hydrogen atom or electron alone. Because the redox pair has a defined reduction potential, it links oxidation of fuels to respiratory chain activity. Many dehydrogenases use NAD+ as a co-substrate and produce NADH. The ratio of NAD+ to NADH reflects metabolic state and influences flux through several pathways.
Beyond redox chemistry, NAD+ acts as a substrate for several enzyme families. ADP-ribosyltransferases, sirtuins, and CD38 ectoenzymes cleave the molecule into nicotinamide and ADP-ribose or related products. These reactions connect NAD+ availability to processes such as DNA repair, chromatin modification, and calcium signaling. Because the coenzyme is used in both electron transfer and signaling, cells maintain separate pools in compartments including the cytosol, mitochondria, and nucleus. The relative sizes and regulation of those pools remain active areas of study.
Cells produce NAD+ through several biosynthetic routes. The salvage pathway recycles nicotinamide, while the Preiss-Handler pathway uses nicotinic acid, and a de novo route can start from tryptophan in some organisms. In mammals, the salvage pathway is generally considered the main source under ordinary conditions. Tissue concentrations vary widely by cell type and compartment, and measured declines with age have been reported in some studies. Whether such changes drive aging or mainly accompany it remains an open question.
Nicotinamide adenine dinucleotide, commonly abbreviated NAD+, is a coenzyme present in all living cells. The molecule consists of two nucleotides linked by phosphate groups, with adenine and a nicotinamide ring as its principal features. In its oxidized form, the nicotinamide ring can accept a hydride ion, becoming NADH. This reversible conversion places NAD+ at the center of many electron-transfer reactions. Its role as a redox carrier is well established across bacteria, plants, fungi, and animals.
Nicotinamide adenine dinucleotide, abbreviated NAD+, is a dinucleotide composed of two nucleotides joined by phosphate groups. One nucleotide contains adenine; the other contains nicotinamide. The molecule exists in oxidized (NAD+) and reduced (NADH) forms, and the reversible hydride transfer between them underlies many metabolic oxidation-reduction reactions. In cells, NAD+ serves as an electron acceptor in pathways such as glycolysis, the citric acid cycle, and oxidative phosphorylation. Its concentration and redox ratio vary by compartment, tissue, and metabolic state.
Beyond redox chemistry, NAD+ is consumed as a substrate by enzymes that transfer ADP-ribose or remove acetyl groups. Sirtuins use NAD+ in deacylation reactions, poly(ADP-ribose) polymerases use it in DNA damage responses, and CD38 hydrolases convert it to signaling metabolites. Because these enzymes compete for the same pool, changes in NAD+ availability can influence multiple cellular processes. The relative contribution of each consumption route differs by cell type and condition, and precise quantitative links remain an active area of study.
In August 2020, British and American officials stated that the Gam-COVID-Vac vaccine would likely be rejected due to concerns that the normally rigorous process of vaccine clinical testing was not followed. As of December 2020, Belarus and Argentina granted emergency use authorization for the vector-based vaccine. On 21 January 2021, Hungary became the first European Union country to register the shot for emergency use, as well as the United Arab Emirates in the Persian Gulf region. On 19 January 2021, the Russian authorities applied for the registration of Sputnik V in the European Union, according to the RDIF. On 10 February, the European Medicines Agency (EMA) said that they had "not received an application for a rolling review or a marketing authorisation for the vaccine". The developers have only expressed their interest that the vaccine be considered for a rolling review, but EMA's Human Medicines Committee (CHMP) and the COVID-19 EMA pandemic Task Force (COVID-ETF) need to give their agreement first before developers can submit their application for initiation of the rolling review process. On 4 March 2021, the Committee for Medicinal Products for Human Use (CHMP) of the EMA started a rolling review of Sputnik V. The EU applicant is R-Pharm Germany GmbH, a subsidiary of the Russian R-Pharm. On 16 June, Reuters reported that approval of Sputnik V will be delayed at least until September because not all the necessary clinical data has been submitted by the deadline.
They wrote in Experientia (later renamed Cellular and Molecular Life Sciences): "Through catalytic actions the macromolecular desoxyribonucleic acids govern the building of macromolecular ribonucleic acids, and, in turn, these control the production of cytoplasmic enzymes. In truth, the enzymic equipment results simultaneously from the effect of ribonucleic acids (catalytic action) and from the effect of substrates (mass action).
=== Regional arms race === After Operation Savannah had failed to prevent the ascension of the MPLA in Angola, the South African political leadership generally accepted that reversing that verdict by force was unrealistic. At the same time, Vorster and Botha had recognised that a total military defeat of PLAN was elusive without the impossible corollary of a victory over the combined FAPLA-PLAN alliance in Angola. Some hardliners in their respective administrations wanted South Africa's full military weight behind Savimbi to help him extinguish the MPLA government, while others favoured simply using it to wage a limited containment exercise against PLAN. An offensive strategy which offered the chance to aggressively attack Angola by land, sea, and air and focus directly on the MPLA's centres of power was never discussed and became more remote as time went on. In its place, therefore, the other popular option was promulgated, which was to focus chiefly on fighting PLAN, the primary threat within the geographical limits of South West Africa proper, and attempting to intimidate Angola in the form of punitive cross-border raids, thus assuming an essentially defensive posture. While Botha never seriously considered the overthrow of the MPLA as a viable objective, he endorsed increasing aid to UNITA for several reasons: it would mend diplomatic relations with the US, especially after the debacle of Operation Argon, UNITA could be molded into a proxy to harass PLAN, and donating captured weapons to Savimbi was cost-effective and deniable.
non-canonical amino acid (ncAA) Also non-standard amino acid. Any amino acid, natural or artificial, that is not one of the 20 or 21 proteinogenic amino acids encoded by the standard genetic code. There are hundreds of such amino acids, many of which have biological functions and are specified by alternative codes or incorporated into proteins accidentally by errors in translation. Many of the best known naturally occurring ncAAs occur as intermediates in the metabolic pathways leading to the standard amino acids, while others have been made synthetically in the laboratory.
=== Cancer === Higher intakes of soy isoflavones may be associated with reduced risks of breast cancer in postmenopausal women and prostate cancer in men. A 2019 systematic review found that intake of soy and soy isoflavones is associated with a lower risk of mortality from gastric, colorectal, breast and lung cancers. The study found that an increase in isoflavone consumption by 10 mg per day was associated with a 7% decrease in risk from all cancers, and an increase in consumption of soy protein by 5 grams per day produced a 12% reduction in breast cancer risk.

=== 6 May === In Russia, the governor of Belgorod Oblast claimed that eight people were killed and 35 others were injured in a Ukrainian drone strike that hit three vehicles in the village of Berezovka. Russia claimed to have taken the villages of Soloviove in Donetsk Oblast and Kotliarivka in Kharkiv Oblast, 20 kilometers from Kupiansk. Ukraine claimed to have destroyed a Russian military fast attack craft in Crimea using a naval drone. Ukrainian forces claimed a series of successful counterattacks against Russian forces near Bakhmut, resulting in an advance of some 500 metres, and inflicting casualties on Russian soldiers and the destruction of four supply trucks, a BMP-2, and a BMD-4 using drones. Russia released footage of a Ka-29 opening fire on a Sea Baby drone armed with a R-73 infrared missile to defend it from helicopters. One missile appeared to have been fired before it was destroyed by gunfire. In response to perceived threats from France, Britain, and the United States, Russia announced it would practice the deployment of tactical nuclear weapons as part of a military exercise.
In pharmacology, bioavailability is a subcategory of absorption and is the fraction (%) of an administered drug that reaches the systemic circulation. By definition, when a medication is administered intravenously, its bioavailability is 100%. However, when a medication is administered via routes other than intravenous, its bioavailability is lower due to intestinal epithelium absorption and first-pass metabolism. Thereby, mathematically, bioavailability equals the ratio of comparing the area under the plasma drug concentration curve versus time (AUC) for the extravascular formulation to the AUC for the intravascular formulation. AUC is used because AUC is proportional to the dose that has entered the systemic circulation. Bioavailability of a drug is an average value; to take population variability into account, deviation range is shown as ±. To ensure that the drug taker who has poor absorption is dosed appropriately, the bottom value of the deviation range is employed to represent real bioavailability and to calculate the drug dose needed for the drug taker to achieve systemic concentrations similar to the intravenous formulation. To dose without knowing the drug taker's absorption rate, the bottom value of the deviation range is used in order to ensure the intended efficacy, unless the drug is associated with a narrow therapeutic window. For dietary supplements, herbs and other nutrients in which the route of administration is nearly always oral, bioavailability generally designates simply the quantity or fraction of the ingested dose that is absorbed.
While there are multiple proposed explanations for how SARS-CoV-2 was introduced into and evolved adaptations suited to human populations, there is significant evidence and agreement that the most likely original viral reservoir for SARS-CoV-2 is horseshoe bats. The closest known viral relatives of SARS-CoV-2 are BANAL-52 and RaTG13, sampled from horseshoe bat droppings in Feuang, Laos, and Yunnan province in China respectively. The evolutionary distance between SARS-CoV-2 and RaTG13 is estimated to be about 50 years (between 38 and 72 years). Bats are a significant reservoir species for a diverse range of coronaviruses, and humans have been found with antibodies for them suggesting that direct infection by bats is common. The zoonotic transmission of SARS-CoV-2 virus to humans took place in the context of exacerbating factors that could make such spillovers more likely. Human contact with bats has increased as human population centers encroach on bat habitats. Several social and environmental factors including climate change, natural ecosystem destruction and wildlife trade have also increased the likelihood for the emergences of zoonosis. One study made with the support of the European Union found climate change increased the likelihood of the pandemic by influencing distribution of bat species. The earliest human cases of SARS-CoV-2 were identified in Wuhan, but the index case remains unknown.
=== 2010–2013: Films with high-profile directors === DiCaprio continued to collaborate with Scorsese in the 2010 psychological thriller film Shutter Island, based on the 2003 novel of the same name by Dennis Lehane. He played Edward "Teddy" Daniels, a U.S. Marshal investigating a psychiatric facility located on an island, who comes to question his own sanity. DiCaprio and Scorsese became interested in the project in 2007, and the former co-produced the film under Appian Way with Phoenix Pictures. Because of the film's disturbing scenes, DiCaprio had nightmares of mass murder during production and considered relaxing with his friends a form of therapy. The film was released to mixed reviews; Peter Bradshaw of The Guardian praised Scorsese's direction and the acting but criticized its twist ending. Peter Travers called it DiCaprio's "most haunting and emotionally complex performance yet", and particularly liked his cave scene with co-star Patricia Clarkson. The film was a commercial success, grossing $294 million worldwide against a budget of $80 million.
== Physiological functions == Through their production of interleukin-22 (IL-22), Th22 cells have been shown to be protective against a number of bacterial and viral pathogens. Interleukin-22 (IL-22) regulates different innate immune mechanisms to eliminate infiltration of invasive pathogens into the skin, gut and respiratory tract. Interleukin-22 (IL-22) influences keratinocytes and epithelial cells to stimulate proliferation and differentiation, thus promoting wound healing and cohesion of barrier integrity. Interleukin-22 (IL-22) also enhances stromal secretion of antimicrobial peptides, as well as production of the chemokines which promote recruitment of other immune cells.

==== FAD ==== Flavin adenine dinucleotide in its oxidized form, FAD is a cofactor of flavoprotein oxidoreductase enzymes. Flavoprotein pyridine nucleotide cytochrome reductases, including FAD catalyse the interchange of reducing equivalents (H+ or electrons). Initial electron donors and final electron acceptors comprise single electron carriers and two electron carrying nicotinamide dinucleotides respectively.
== Development and biochemistry of the Strep-tag == Streptavidin is a tetrameric protein expressed in Streptomyces avidinii. Because of Streptavidin's high affinity for vitamin H (biotin), Streptavidin is commonly used in the fields of molecular biology and biotechnology. The Strep-tag was originally selected from a genetic library to specifically bind to a proteolytically truncated "core" version of streptavidin. Over the years, the Strep-tag was systemically optimized, to permit a greater flexibility in the choice of attachment site. Further, its interaction partner, Streptavidin, was also optimized to increase peptide-binding capacity, which resulted in the development of Strep-Tactin. The binding affinity of Strep-tag to Strep-Tactin is nearly 100 times higher than from Strep-tag to Streptavidin. The so-called Strep-tag system, consisting of Strep-tag and Strep-Tactin, has proven particularly useful for the functional isolation and analysis of protein complexes in proteome research.
MH+(H2O)m → MH+ + mH2O If the mobile phase contains solvents with a higher proton affinity than water, proton-transfer reactions take place that lead to protonated the solvent with higher proton affinity. For example, when methanol solvent is present, the cluster solvent ions would be CH3OH2+(H2O)n(CH3OH)m. Fragmentation does not normally occur inside the APCI source. If a fragment ion of a sample is observed, thermal degradation has taken place by the heated nebulizer interface, followed by the ionization of the decomposition products. In a major distinction from chemical ionization, the electrons needed for the primary ionization are not produced by a heated filament, as a heated filament cannot be used under atmospheric pressure conditions. Instead, the ionization must occur using either corona discharges or β- particle emitters, which are both electron sources capable of handling the presence of corrosive or oxidizing gases.
Sucrose esters or sucrose fatty acid esters are a group of non-naturally occurring surfactants chemically synthesized from the esterification of sucrose and fatty acids (or glycerides). This group of substances is remarkable for the wide range of hydrophilic-lipophilic balance (HLB) that it covers. The polar sucrose moiety serves as a hydrophilic end of the molecule, while the long fatty acid chain serves as a lipophilic end of the molecule. Due to this amphipathic property, sucrose esters act as emulsifiers; i.e., they have the ability to bind both water and oil simultaneously. Depending on the HLB value, some can be used as water-in-oil emulsifiers, and some as oil-in-water emulsifiers. Sucrose esters are used in cosmetics, food preservatives, food additives, and other products. A class of sucrose esters with highly substituted hydroxyl groups, olestra, is also used as a fat replacer in food.

NAD+ is the oxidized form, while NADH is the reduced form carrying an additional hydride equivalent. The pair participates in reversible electron transfer reactions. Their ratio helps indicate the redox state of a compartment.
NAD+ itself is not classified as a vitamin, but its precursor niacin is an essential nutrient in humans. Cells synthesize NAD+ from niacin, nicotinamide, nicotinamide riboside, or tryptophan. The intact dinucleotide is not obtained directly from typical diets in meaningful amounts.
Age-related studies often examine whether NAD+ levels decline in tissues and whether that decline affects mitochondrial function or DNA repair. Interventions using precursor molecules raise open questions about cause and effect. Current evidence does not establish that changing NAD+ levels slows human aging.
NAD+ is an oxidized dinucleotide coenzyme that carries electrons in metabolic reactions. It is also consumed by signaling enzymes, including sirtuins and PARPs. Its reduced form is NADH.
