In glycolysis, NAD+ accepts electrons during the oxidation of glyceraldehyde-3-phosphate, forming NADH. The tricarboxylic acid cycle and fatty acid oxidation also generate NADH, which donates electrons to the mitochondrial electron transport chain. This flow supports ATP synthesis and helps maintain the redox balance of the cell. Other dehydrogenases use NAD+ as a cofactor for biosynthetic reductions and detoxification reactions. NADH is later reoxidized to sustain continued flux through these pathways.
Beyond electron transfer, NAD+ serves as a substrate for enzymes that cleave the molecule and transfer ADP-ribose units. Sirtuins, poly(ADP-ribose) polymerases, and CD38 consume NAD+ in regulatory reactions. These activities link NAD+ availability to DNA repair, chromatin modification, calcium signaling, and metabolic stress responses. Because consumption can exceed biosynthesis under some conditions, cellular NAD+ levels are dynamic rather than fixed. Enzyme affinity and local synthesis also influence how much NAD+ is available for signaling.
NAD+ is a dinucleotide composed of adenine, ribose, and nicotinamide moieties linked by phosphate groups. Its oxidized form carries a positive charge on the nicotinamide ring, which enables reversible hydride transfer. The molecule functions as a coenzyme in oxidoreductase reactions rather than as a dietary vitamin in its intact form. Cells maintain separate pools in cytoplasm, mitochondria, and nucleus. This compartmentalization allows distinct redox environments while preserving a shared chemical identity.
Commercial NAD+ is supplied as a solid, often as the free acid or a salt, and purity is verified by chromatographic methods. Laboratories typically store it desiccated at minus 20 degrees Celsius or below. Working solutions are prepared fresh because even sterile aqueous solutions can lose activity over hours to days depending on pH and temperature. Documentation may include a certificate of analysis, an assay value, and a recommended retest date. Researchers should verify identity and purity when results depend on precise cofactor concentrations.
NAD+ is commonly measured by high-performance liquid chromatography with ultraviolet detection, often at 254 or 260 nm. Enzymatic cycling assays provide higher sensitivity by coupling NAD+ to a reporter reaction. Mass spectrometry can distinguish NAD+ from close analogues and confirm isotope labeling. Sample preparation usually involves rapid quenching of metabolism to prevent interconversion with NADH. Because NAD+ and NADH differ by one hydride, extraction conditions strongly affect the measured ratio.

Research on NAD+ often examines changes with age, diet, exercise, and disease states, but causal relationships are difficult to establish. Some studies measure NAD+ levels, while others assess enzyme activity or downstream markers. In the literature, terms such as "NAD+ decline" and "NAD+ boosting" appear in both scientific and commercial contexts, sometimes without precise definitions. Whether changes in measured NAD+ directly produce health effects remains an open question. Results from cells, animals, and humans cannot be assumed to translate directly.
Measuring NAD+ in biological samples requires rapid processing because the compound can degrade or interconvert after collection. Common approaches include enzymatic cycling assays, high-performance liquid chromatography, and mass spectrometry. Each method has different sensitivity, specificity, and susceptibility to interference from related nucleotides. Sample type matters: cultured cells, animal tissues, and human blood present distinct challenges. Reported values can vary widely across laboratories because of differences in extraction, normalization, and analytical platform. Standardization remains an open issue in the field.
Beyond electron transfer, NAD+ serves as a substrate for enzymes that cleave it and attach its ADP-ribose portion to other molecules. This group includes poly(ADP-ribose) polymerases, CD38, and sirtuins. Such reactions consume NAD+ and can influence its availability for metabolism. Cells replenish NAD+ through a salvage pathway that recycles nicotinamide and through routes starting from tryptophan or vitamin B3 forms. How these synthesis and consumption routes are coordinated across tissues remains an active area of study, and compartment-specific concentrations are difficult to measure directly.
Nicotinamide adenine dinucleotide, abbreviated NAD+, is a dinucleotide built from adenine, nicotinamide, two ribose sugars, and two phosphate groups. The oxidized form carries a positive charge on the nicotinamide ring, while the reduced form, NADH, is neutral. This pair acts as a reversible electron carrier in cells. NAD+ is present in bacteria, plants, animals, and fungi. Its structure allows it to accept and donate electrons without being consumed in the reactions it supports.
In redox reactions, NAD+ accepts a hydride ion, which consists of two electrons and one proton. The hydride adds to the nicotinamide ring at a specific carbon, converting NAD+ into NADH. Dehydrogenase enzymes use this step in glycolysis, the citric acid cycle, and fatty acid oxidation. NADH later donates electrons to the mitochondrial electron transport chain, helping to drive ATP synthesis. The balance between NAD+ and NADH reflects the metabolic state of a cell, and shifts in that balance can alter how pathways operate.
The stability of NAD+ depends on pH, temperature, light exposure, and the presence of degradative enzymes. Aqueous solutions are generally more stable under mildly acidic to neutral conditions and degrade faster under alkaline conditions or prolonged heat. The solid is hygroscopic and should be stored desiccated, often frozen, and protected from repeated freeze-thaw cycles. In laboratory handling, aliquots reduce repeated temperature changes, and chelating agents may limit metal-catalyzed hydrolysis in some buffers. These practices matter because even small amounts of NADH or hydrolysis products can interfere with quantitative assays.
Quality control for NAD+ materials typically combines identity, purity, and water content checks. Identity may be confirmed by ultraviolet spectrum, retention time in chromatography, or mass accuracy, while purity is assessed by HPLC peak area or quantitative nuclear magnetic resonance. Residual water and solvents can affect molar calculations and enzyme assays, so Karl Fischer titration or thermogravimetric analysis may be used. Commercial materials vary in grade and counterion form, and published methods should specify the exact salt or hydrate when reporting concentrations. Regulatory status depends on intended use, with research reagents, dietary ingredients, and clinical products treated under different frameworks.
Quantification of NAD+ in biological samples usually relies on separation techniques coupled to sensitive detection. High-performance liquid chromatography with ultraviolet detection can measure the oxidized form by its absorbance near 260 nm, while mass spectrometry provides greater specificity and can distinguish NAD+ from close analogs. Enzymatic cycling assays use coupled dehydrogenase reactions to amplify signal and estimate NAD+ concentrations in cell or tissue extracts. Because NAD+ and NADH interconvert rapidly, sample preparation must quench metabolism quickly and preserve the redox state before analysis.
Nicotinamide adenine dinucleotide, abbreviated NAD+, is a dinucleotide composed of two nucleotides joined by phosphate groups. One nucleotide contains adenine; the other contains nicotinamide. The molecule exists in oxidized (NAD+) and reduced (NADH) forms, and the reversible hydride transfer between them underlies many metabolic oxidation-reduction reactions. In cells, NAD+ serves as an electron acceptor in pathways such as glycolysis, the citric acid cycle, and oxidative phosphorylation. Its concentration and redox ratio vary by compartment, tissue, and metabolic state.
Beyond redox chemistry, NAD+ is consumed as a substrate by enzymes that transfer ADP-ribose or remove acetyl groups. Sirtuins use NAD+ in deacylation reactions, poly(ADP-ribose) polymerases use it in DNA damage responses, and CD38 hydrolases convert it to signaling metabolites. Because these enzymes compete for the same pool, changes in NAD+ availability can influence multiple cellular processes. The relative contribution of each consumption route differs by cell type and condition, and precise quantitative links remain an active area of study.
=== Economics === In 2007, paroxetine was ranked 94th on the list of bestselling drugs, with over $1 billion in sales. In 2006, paroxetine was the fifth-most prescribed antidepressant in the U.S. retail market, with more than 19.7 million prescriptions. In 2007, sales had dropped slightly to 18.1 million but paroxetine remained the fifth-most prescribed antidepressant in the U.S.
==== Therapeutic treatments ==== D-amino acid oxidase is used in therapeutic treatments such as regulation of hormones, regulation of hypertension, treatment of schizophrenia, treatment of psychiatric and cognitive disorders, and possible pain reduction. Changing the amount of D-amino acid oxidase transporters with the use of drugs has therapeutic effects on schizophrenia. D-amino acid oxidase regulates D-aspartate, which regulates the secretion of melatonin, prolactin, testosterone, luteinizing hormone and growth hormone. By regulating D-amino acid oxidase, D-aspartate can also be regulated and control hormone secretion. Increased D-amino acid oxidase activity has been correlated with psychiatric and cognitive disorders, so reducing D-amino acid oxidase can have therapeutic effects on these disorders. D-amino acid oxidase helps produce L-6-hydroxynorleucine, which then generates omapatrilat. Omapatrilat inhibits angiotensin-converting enzyme and neutral endopeptidase and effectively reduces hypertension. D-amino acid oxidase also may have an effect on pain stimuli, but it is not confirmed yet.
==== Butyrophenone(s) ==== Lumateperone (Caplyta) – In December 2019, lumateperone, a presynaptic D2 receptor partial agonist and postsynaptic D2 receptor antagonist, received its first global approval in the US for the treatment of schizophrenia in adults. In 2020 and 2021 FDA approved for depressive episodes associated with bipolar I or II disorder in adults, as monotherapy and as adjunctive therapy with lithium or valproate.
== Properties and structure == Thiophene is a colorless liquid at room temperature. The high reactivity of thiophene toward sulfonation is the basis for the separation of thiophene from benzene, which are difficult to separate by distillation due to their similar boiling points (4 °C difference at ambient pressure). Like benzene, thiophene forms an azeotrope with ethanol. The molecule is flat; the bond angle at the sulfur is around 93°, the C–C–S angle is around 109°, and the other two carbons have a bond angle around 114°. The C–C bonds to the carbons adjacent to the sulfur are about 1.34 Å, the C–S bond length is around 1.70 Å, and the other C–C bond is about 1.41 Å.
== Mechanism == Classified as a metalloexopeptidase, carboxypeptidase A consists of a single polypeptide chain bound to a zinc ion. This characteristic metal ion is located within the active site of the enzyme, along with five amino acid residues that are involved in substrate binding: Arg-71, Arg-127, Asn-144, Arg-145, Tyr-248, and Glu-270. X-ray crystallographic studies have revealed five subsites on the protein. These allosteric sites are involved in creating the ligand-enzyme specificity seen in most bioactive enzymes. One of these subsites induces a conformational change at Tyr-248 upon binding of a substrate molecule at the primary active site. The phenolic hydroxyl of tyrosine forms a hydrogen bond with the terminal carboxylate of the ligand. In addition, a second hydrogen bond is formed between the tyrosine and a peptide linkage of longer peptide substrates. These changes make the bond between the enzyme and ligand, whether it is substrate or inhibitor, much stronger. This property of carboxypeptidase A led to the first clause of Daniel E. Koshland, Jr.’s “induced fit” hypothesis. The S1 sub-site is where catalysis occurs in CPA, and the zinc ion is coordinated by Glu-72, His-69, and His-196 enzyme residues. A plane exists that bisects the active-site groove where residues Glu-270 and Arg-127 are on opposite sides of the zinc-water coupled complex. The zinc is electron rich due to glutamine ligands coordinating the zinc because before substrate binds, Glu-72 coordinates bidentate but shifts to monodentate after substrate binds.

7-Eleven is the largest convenience store chain in Taiwan, and is owned by President Chain Store Corporation (PCSC). The first fourteen stores opened in 1979, and struggled to make a profit. Southland Corporation partnered with Uni-President to modernize the stores. However, business was still slow, and Uni-President opted to stock Asian foods. In 1986, 7-Eleven made its first profit in Taiwan. The 5,000th store was opened in July 2014. In January 2018, an experimental and unstaffed shop branded the X-Store was opened. 7-Eleven announced plans to operate a combination store in partnership with Domino's Pizza in February 2019. The 6,000th store was opened on February 20, 2021. The 7,000th store was opened on July 5, 2024.
=== Preservation and storage === Specimens are typically housed in light-, water-, and insect-proof cabinets within herbaria or fungaria. After collection, fungal specimens are accessioned and their data recorded, then processed, labelled, mounted, and filed. Preparation commonly includes prompt drying by warm air, silica gel, or freeze drying; large or watery sporocarps may be sliced to hasten drying. Field labels and notes record the collector, date, locality, habitat, substratum or host, and fresh characters, and incoming material is checked for dryness and often disinfested by deep freezing before being packeted or boxed and filed within the collection. Long-term storage ideally relies on climate-controlled conditions, because warm, humid environments increase the risks of mould growth and insect attack. Routine pest management can include monitoring and freezing incoming specimens before they enter the main collection. In some large collections, high-density compactor systems with open-faced cabinets have been used to increase storage capacity two- to threefold over conventional cabinet arrangements while still allowing the closed units to be sealed against insect pests. Storage methods have varied over time and between institutions. Historical practice included storing flat specimens in folded paper packets attached to herbarium sheets, while bulkier material was often kept in boxes sized to fit the filing system.
Automated insulin delivery systems are automated (or semi-automated) systems designed to assist people with insulin-requiring diabetes, by automatically adjusting insulin delivery in response to blood glucose levels. Currently available systems (as of October 2020) can only deliver (and regulate delivery of) a single hormone—insulin. Other systems currently in development aim to improve on current systems by adding one or more additional hormones that can be delivered as needed, providing something closer to the endocrine functionality of the pancreas. The endocrine functionality of the pancreas is provided by islet cells which produce the hormones insulin and glucagon. Artificial pancreatic technology mimics the secretion of these hormones into the bloodstream in response to the body's changing blood glucose levels. Maintaining balanced blood sugar levels is crucial to the function of the brain, liver, and kidneys. Therefore, for people with diabetes, it is necessary that the levels be kept balanced when the body cannot produce insulin itself. Automated insulin delivery (AID) systems are often referred to using the term artificial pancreas, but the term has no precise, universally accepted definition. For uses other than automated insulin delivery, see Artificial pancreas (disambiguation).
Staphylococcus, Streptococcus and diphtheria bacillus (Corynebacterium diphtheriae) were easily killed, but there was no effect on typhoid bacterium (Salmonella typhimurium) and the bacterium once thought to cause influenza (Haemophilus influenzae). He prepared a culture method from which he could obtain the mould juice, which he called "penicillin" on 7 March 1929, "to avoid the repetition of the rather cumbersome phrase 'mould broth filtrate'." In his Nobel lecture of 1945 he gave a further explanation, saying:

=== Economic situation === Because of their wide needs, the overall chelating agents growth was 4% annually during 2009–2014 and the trend is likely to increase. Aminopolycarboxylic acids chelators are the most widely consumed chelating agents; however, the percentage of the greener alternative chelators in this category continues to grow. The consumption of traditional aminopolycarboxylates chelators, in particular the EDTA (ethylenediaminetetraacetic acid) and NTA (nitrilotriacetic acid), is declining (−6% annually), because of the persisting concerns over their toxicity and negative environmental impact. In 2013, these greener alternative chelants represented approximately 15% of the total aminopolycarboxylic acids demand. This is expected to rise to around 21% by 2018, replacing and aminophosphonic acids used in cleaning applications. Examples of some Greener alternative chelating agents include ethylenediamine disuccinic acid (EDDS), polyaspartic acid (PASA), methylglycinediacetic acid (MGDA), glutamic diacetic acid (L-GLDA), citrate, gluconic acid, amino acids, plant extracts etc.
=== Pre-industrial technology === Nanoparticles were used by artisans since prehistory, albeit without knowledge of their nature. They were used by glassmakers and potters in Classical Antiquity, as exemplified by the Roman Lycurgus cup of dichroic glass (4th century CE) and the lusterware pottery of Mesopotamia (9th century CE). The latter is characterized by silver and copper nanoparticles dispersed in the glassy glaze.
While the federal constitution assigns local authorities to the exclusive jurisdiction of state governments, in practice, the federal Ministry of Housing and Local Government oversees the regulation of local laws and policies. Mayors (or presidents for municipal and district councils) and councillors are appointed by the respective state governments, or in the case of the federal territories, by the federal government. Federal laws assign land matters, including the delineation of districts, to the purview of state governments. Except Perlis and the federal territories, each state is divided into districts, which are further subdivided into mukims. In Sabah and Sarawak, districts are grouped into divisions. In contrast to local governments that manage municipal administration and infrastructure development, districts are solely utilised for land taxation. The 13 states are based on historical Malay kingdoms, and 9 of the 11 Peninsular states, known as the Malay states, retain their royal families. The King is elected by and from the nine rulers to serve a five-year term. This King appoints governors serving a four-year term for the states without monarchies, after consultations with the chief minister of that state. Each state has its written constitution. Sabah and Sarawak have considerably more autonomy than the other states, most notably having separate immigration policies and controls, and unique residency status.
== Treatment == Management of chronic granulomatous disease revolves around two goals: 1) diagnose the disease early so that antibiotic prophylaxis can be given to keep an infection from occurring, and 2) educate the patient about his or her condition so that prompt treatment can be given if an infection occurs.

NAD+ is the oxidized form, while NADH is the reduced form carrying an additional hydride equivalent. The pair participates in reversible electron transfer reactions. Their ratio helps indicate the redox state of a compartment.
NAD+ itself is not classified as a vitamin, but its precursor niacin is an essential nutrient in humans. Cells synthesize NAD+ from niacin, nicotinamide, nicotinamide riboside, or tryptophan. The intact dinucleotide is not obtained directly from typical diets in meaningful amounts.
Age-related studies often examine whether NAD+ levels decline in tissues and whether that decline affects mitochondrial function or DNA repair. Interventions using precursor molecules raise open questions about cause and effect. Current evidence does not establish that changing NAD+ levels slows human aging.
NAD+ and NADH can interconvert quickly after a sample is collected, which can alter the measured ratio. Rapid quenching and cold handling limit enzymatic and chemical changes.
