NAD+ is the oxidized form of nicotinamide adenine dinucleotide, a coenzyme built from two nucleotides joined by a phosphate linkage. One nucleotide carries adenine, and the other carries nicotinamide; the plus sign denotes a formal positive charge on the nicotinamide ring, not a free proton. In cells, NAD+ and its reduced partner NADH form a reversible redox pair. That pair participates in electron transfer reactions throughout metabolism. The abbreviation NAD+ is common in biochemistry, while NAD(H) sometimes denotes the combined pool.
The molecule was first described in the early twentieth century as a factor that promoted fermentation in yeast extracts. Later work linked it to hydrogen transfer and to the oxidation of nutrients in living tissues. Its structure was resolved as a dinucleotide, which explained why it could accept and donate electrons at specific enzyme sites. Today, NAD+ is recognized as a central substrate and signaling precursor, not merely a metabolic cofactor. Whether all observed NAD+ changes reflect causal signaling remains an open question.
Related compounds include NADH, the reduced form, and NADP+, which carries an additional phosphate group. NADP+ and NADPH often serve in biosynthetic and antioxidant reactions, while NAD+ and NADH are more associated with energy-yielding catabolism. Nicotinamide, nicotinic acid, and nicotinamide riboside are precursors that can enter salvage pathways. The exact contribution of dietary precursors to tissue NAD+ pools is an area of active investigation. Some studies measure labeled precursors to trace those routes.
Quantification of NAD+ in biological samples usually relies on separation techniques coupled to sensitive detection. High-performance liquid chromatography with ultraviolet detection can measure the oxidized form by its absorbance near 260 nm, while mass spectrometry provides greater specificity and can distinguish NAD+ from close analogs. Enzymatic cycling assays use coupled dehydrogenase reactions to amplify signal and estimate NAD+ concentrations in cell or tissue extracts. Because NAD+ and NADH interconvert rapidly, sample preparation must quench metabolism quickly and preserve the redox state before analysis.
The stability of NAD+ depends on pH, temperature, light exposure, and the presence of degradative enzymes. Aqueous solutions are generally more stable under mildly acidic to neutral conditions and degrade faster under alkaline conditions or prolonged heat. The solid is hygroscopic and should be stored desiccated, often frozen, and protected from repeated freeze-thaw cycles. In laboratory handling, aliquots reduce repeated temperature changes, and chelating agents may limit metal-catalyzed hydrolysis in some buffers. These practices matter because even small amounts of NADH or hydrolysis products can interfere with quantitative assays.
Quality control for NAD+ materials typically combines identity, purity, and water content checks. Identity may be confirmed by ultraviolet spectrum, retention time in chromatography, or mass accuracy, while purity is assessed by HPLC peak area or quantitative nuclear magnetic resonance. Residual water and solvents can affect molar calculations and enzyme assays, so Karl Fischer titration or thermogravimetric analysis may be used. Commercial materials vary in grade and counterion form, and published methods should specify the exact salt or hydrate when reporting concentrations. Regulatory status depends on intended use, with research reagents, dietary ingredients, and clinical products treated under different frameworks.

Beyond redox chemistry, NAD+ serves as a substrate for enzymes that transfer ADP-ribose or remove acetyl groups. Sirtuins, PARPs, and CD38-family enzymes consume NAD+ and produce nicotinamide and ADP-ribose-related products. These reactions link NAD+ availability to DNA repair, chromatin modification, and cellular signaling. Because the molecule is central to energy metabolism and regulation, changes in its concentration are studied in aging, immunity, and metabolic research. The balance between synthesis and consumption varies by tissue, developmental stage, and physiological state.
In humans, NAD+ can be synthesized from nicotinic acid, nicotinamide, nicotinamide riboside, and tryptophan through overlapping pathways. The salvage pathway recycles nicotinamide back to NAD+ and is often considered a major route in many tissues. Dietary precursors and intracellular recycling both contribute to the pool, but the quantitative importance of each source remains an active research question. NAD+ levels are not uniform across organs or cell compartments. Measurements in blood do not necessarily reflect concentrations inside tissues.
Beyond electron transfer, NAD+ serves as a substrate for enzymes that cleave it and attach its ADP-ribose portion to other molecules. This group includes poly(ADP-ribose) polymerases, CD38, and sirtuins. Such reactions consume NAD+ and can influence its availability for metabolism. Cells replenish NAD+ through a salvage pathway that recycles nicotinamide and through routes starting from tryptophan or vitamin B3 forms. How these synthesis and consumption routes are coordinated across tissues remains an active area of study, and compartment-specific concentrations are difficult to measure directly.
Nicotinamide adenine dinucleotide, abbreviated NAD+, is a dinucleotide built from adenine, nicotinamide, two ribose sugars, and two phosphate groups. The oxidized form carries a positive charge on the nicotinamide ring, while the reduced form, NADH, is neutral. This pair acts as a reversible electron carrier in cells. NAD+ is present in bacteria, plants, animals, and fungi. Its structure allows it to accept and donate electrons without being consumed in the reactions it supports.
In redox reactions, NAD+ accepts a hydride ion, which consists of two electrons and one proton. The hydride adds to the nicotinamide ring at a specific carbon, converting NAD+ into NADH. Dehydrogenase enzymes use this step in glycolysis, the citric acid cycle, and fatty acid oxidation. NADH later donates electrons to the mitochondrial electron transport chain, helping to drive ATP synthesis. The balance between NAD+ and NADH reflects the metabolic state of a cell, and shifts in that balance can alter how pathways operate.

The smallpox vaccine has also proven useful in preventing emergent infectious diseases caused by closely-related orthopoxviruses such as Mpox. Three specific smallpox vaccines have been approved in at least one country for Mpox prevention: MVA-BN, LC16m8, and ACAM2000. The Mpox vaccine is on the World Health Organization's List of Essential Medicines.
=== 1967–1968: RoBee's and Marriott === In 1967, the Azar's Big Boy restaurant franchise started RoBee's House of Beef restaurants in Ft. Wayne, Indiana. The Marriott Corp., which had acquired Bob's Big Boy and the Big Boy trademark in 1967, acquired RoBee's in February 1968 with plans to expand nationwide. RoBee's franchises would first be offered to Big Boy franchisees to coincide with their existing Big Boy territory. At the time there were 13 RoBee's restaurants in six states. During the acquisition, in January 1968, the competing roast beef chain Arby's sued RoBee's for trademark infringement and (other similarities that it considered) unfair competition. Because "RoBee's" sounded too much like "Arby's" the settlement required a new brand name and Marriott wanted something recognizable. Big Boy founder Bob Wian, then sitting on Marriott's board of directors, was friends with Roy Rogers' agent and suggested that the company approach Rogers about the use of his name. Already interested in associating with a chain restaurant, Rogers was in similar discussions with another company when Marriott called. Nonetheless, he accepted Marriott's offer: Rogers would receive a licensing fee for use of his name and also be paid for personal appearances at the restaurants. The restaurants would be called "Roy Rogers Roast Beef Sandwich" restaurants, and despite Arby's complaints, it retained RoBee's building design and covered wagon logo design.

=== Chemical === Neptunium has five ionic oxidation states ranging from +3 to +7 when forming chemical compounds, which can be simultaneously observed in solutions. It is the heaviest actinide that can lose all its valence electrons in a stable compound. The most stable state in solution is +5, but the valence +4 is preferred in solid neptunium compounds. Neptunium metal is very reactive. Ions of neptunium are prone to hydrolysis and formation of coordination compounds.
Another improvement would be a transplantation of the pancreas or beta cell to avoid periodic insulin administration. This would result in a self-regulating insulin source. Transplantation of an entire pancreas (as an individual organ) is difficult and relatively uncommon. It is often performed in conjunction with liver or kidney transplant, although it can be done by itself. It is also possible to do a transplantation of only the pancreatic beta cells. However, islet transplants had been highly experimental for many years, but some researchers in Alberta, Canada, have developed techniques with a high initial success rate (about 90% in one group). Nearly half of those who got an islet cell transplant were insulin-free one year after the operation; by the end of the second year that number drops to about one in seven. However, researchers at the University of Illinois at Chicago (UIC) have slightly modified the Edmonton Protocol procedure for islet cell transplantation and achieved insulin independence in diabetic people, with fewer but better-functioning pancreatic islet cells. Beta cell transplant may become practical. Additionally, some researchers have explored the possibility of transplanting genetically engineered non-beta cells to secrete insulin.
The report stated that "continued destruction which came after the ceasefire agreement was not justifiable by any means" and noted that since "extended Russian military action reaching out into Georgia was conducted in violation of international law, Georgian military forces were acting in legitimate self-defence under Article 51 of the UN Charter." Although further Russian military advances into the Georgian territories were explained by the Russian authorities as necessitated to avert possible Georgian counter-attacks from the Gori Municipality and to avert "imminent Georgian attack on Abkhazia", the commission noted that "The Georgian armed forces were hardly ever able to conduct military operations on two fronts at the same time", and "In practical terms, there were no Georgian combat troops in western Georgia when the Russian operation there started". The commission noted, "Georgia did not use force against Russian troops on Russian territory, but only on Georgian territory." The commission stated that an attack by Georgian troops on Russian peacekeepers present in Georgia – "if not in self-defence against a Russian attack", would not be justified. The commission wrote that it was "not entirely clear that Georgian military action against the base was aimed specifically at Russia".
== Use and management == Drains help to remove contents, usually fluids, from inside the body. This is beneficial since fluid accumulation may cause distension and pressure, which can lead to pain. For example, nasogastric (NG) tubes inserted through the nose and into the stomach can help remove stomach contents for patients who have a blockage further along in their gastrointestinal tract. After surgery, drains can be placed to remove blood, lymph, or other fluids that accumulate in the wound bed. This helps to promote wound healing and allows healthcare providers to monitor the wound for any signs of internal infection or damage to surgically repaired structures. Drains may be classified as passive or active, open or closed, and external or internal. Passive drains rely on gravity or capillary action to remove fluid, whereas active drains rely on a suction/vacuum force, whether that be through connection to wall suction, a portable suction device, or a bulb that has been squeezed to create a vacuum. Open drains are commonly used for superficial wounds and drain into dressings or a stoma bag. Closed drains are tubes or other channel-like structures that are connected to a container, thereby creating a closed system. External drains go from inside the body to outside the body and can be seen, while internal drains are completely inside the body. An example of an internal drain is a ventriculo-peritoneal shunt, which is a tube that connects ventricles of the brain to the peritoneal cavity. This helps remove extra cerebrospinal fluid from the brain.

The Straw Hats' Jolly Roger was first used by protesters in Indonesia as early as October 2023 in Yogyakarta, during protests against the Gaza genocide. Student protesters in Indonesia begun flying the flag again in February 2025. Truck drivers began flying the Straw Hat Pirates' flag in late July 2025 to protest the prohibition of ODOL (over dimension, overload) trucks throughout several cities in Java, ongoing since 19 June 2025. The flag came into prominence during the 2025 Indonesian anti-government protests which started in the weeks leading up to Independence Day on 17 August. In July, President Prabowo Subianto had called on Indonesians to fly the red and white national flag to celebrate the 80th anniversary of the country's independence. Following the president's speech an apparel store received "thousands of orders" for the Jolly Roger flag, because "The anime reflects the injustice and inequality that Indonesians experience".
=== Oceania === The Lapita culture, active across a large area of the Pacific Ocean around 1000 BC, made widespread use of obsidian tools and engaged in long distance obsidian trading. The complexity of the production technique for these tools, and the care taken in their storage, may indicate that beyond their practical use they were associated with prestige or high status. Obsidian was also used on Rapa Nui (Easter Island) for edged tools such as mataia and the pupils of the eyes of their Moai (statues), which were encircled by rings of bird bone. Obsidian was used to inscribe the Rongorongo glyphs.
Fuel savings: According to the GITI, for every 10% of under-inflation on each tire on a vehicle, a 1% reduction in fuel economy will occur. In the United States alone, the Department of Transportation estimates that under inflated tires waste 2 billion US gallons (7,600,000 m3) of fuel each year. Extended tire life: Under inflated tires are the number one cause of tire failure and contribute to tire disintegration, heat buildup, ply separation and sidewall/casing breakdowns. Further, a difference of 10 pounds per square inch (69 kPa; 0.69 bar) in pressure on a set of duals literally drags the lower pressured tire 2.5 metres per kilometre (13 feet per mile). Moreover, running a tire even briefly on inadequate pressure breaks down the casing and prevents the ability to retread. Not all sudden tire failures are caused by under-inflation. Structural damages caused, for example, by hitting sharp curbs or potholes, can also lead to sudden tire failures, even a certain time after the damaging incident. These cannot be proactively detected by any TPMS. Improved safety: Under-inflated tires lead to tread separation and tire failure, resulting in 40,000 accidents, 33,000 injuries and over 650 deaths per year. Properly inflated tires provide greater stability, handling, and braking efficiency. Environmental efficiency: Under-inflated tires, as estimated by the US Department of Transportation, release over 26 billion kilograms (57.5 billion pounds) of unnecessary carbon-monoxide pollutants into the atmosphere each year in the United States alone.
=== Transportation choices === Many members of Generation Z no longer view car ownership as a status symbol, a life milestone, or a ticket to freedom. However, large swaths of this cohort have not had a driver's license or owned a car because of their economic situation rather than their concerns for the environment or dislike of cars. Moreover, decades of auto-centric urban development in the United States have led to under-investment in walkable neighborhoods, bicycle lanes, and public transit. The number of Generation Z consumers taking out auto loans was rising drastically during the late 2010s. While a significant portion of Generation Z consider environmental friendliness to be an important factor, more than their predecessors when they were at the same age, affordability was much for important to them. Generation Z is not particularly concerned with style or brand; they are more interested in safety. They tend to be more receptive towards self-driving cars. They are also interested in the interior electronic technology of the cars they might purchase. More specifically, they would like to be able to hook up their smartphones to the Bluetooth-capable audio systems and backup cameras.
== Subcellular distribution == The peptide recognition domain of epiplakin is able to bind to keratins in vitro; however, in cells the epiplakin associates with keratin intermediate filaments networks only under conditions in which cellular stress is absent. Otherwise, epiplakin remains universally cytoplasmic and not bound to the intermediate filaments.

It indicates a formal positive charge on the nicotinamide ring. The molecule is not simply a protonated acid, and the charge is part of its redox chemistry.
NAD+ is the oxidized form, while NADH is the reduced form carrying two additional electrons and a proton. The two forms interconvert in many metabolic reactions.
No. NADP+ contains an extra phosphate group on the adenine ribose. NADP+ and NADPH tend to participate in different biosynthetic and antioxidant pathways.
Common methods include LC-MS, HPLC with UV detection, and enzymatic cycling assays. Rapid quenching is needed because NAD+ and NADH interconvert. The chosen method should be validated for the sample matrix.
